چکیده

Cells of Bacillus megaterium can use the externally added precursors UDP-N-acetyl-D-glucosamine and UDP-N-acetylD mU!‘aIUyl L danyl D ghItamy1 mes o diaminopimelyl D alanyl-n-alanine to synthesize peptidoglycan if the bacteria were first treated with toluene. Toluene-treated bacteria could polymerize these precursors into peptidoglycan which was attached to previously existing cell wall by peptide crossbridges.

Reconstitution of cell wall synthesis in toluene- and LiCl-treated Bacillus megaterium cells by addition of a soluble protein extract.

Proteins required for the synthesis of peptidoglycan and incorporation of diaminopimelic acid into cell walls have been solubilized from Bacillus megaterium toluene-treated cells. Some of these proteins might have been removed from the cytoplasmic membrane through the cell wall by extraction with LiCl. The solubilized proteins have molecular weights in the range of 40,000 to 70,000 and can be a...

Purification and properties of a protein factor stimulating peptidoglycan synthesis in toluene- and Licl-treated Bacillus megaterium cells.

A protein factor, called PG-I, can be solubilized from toluene-treated Bacillus megaterium cells by LiCl extraction. After LiCl extraction, peptidoglycan synthesis by the toluene-treated cells is decreased. Protein PG-I can be added back to the extracted cells to stimulate peptidoglycan synthesis. This factor has now been purified 124-fold. It has a molecular weight of 42,000 as estimated by Se...

Biosynthesis of peptidoglycan in the one million molecular weight range by membrane preparations from Bacillus megaterium.

Peptidoglycan made in vifro by membrane preparations of Bacillus megaterium had a buoyant density in CsCl different from that of native cell wall. This newly made material was not covalently attached to substantial amounts of previously formed cell material such as wall. From sedimentation velocity centrifugation, the molecular weight of much of the in vitro product was found to be greater than...

Purification of the peptidoglycan transglycosylase of Bacillus megaterium.

The peptidoglycan transglycosylase of Bacillus megaterium has been purified approximately 500-fold from a crude membrane fraction. This protein is likely to be the one previously called PG-II and was assayed by its ability to reconstitute with a crude phospho-N-acetyl-muramyl-pentapeptide translocase preparation and partially purified N-acetylglucosaminyl transferase to give peptidoglycan synth...

Biosynthesis of the Peptidoglycan of Bacterial Cell Walls

An uncross-linked monomer (nascent peptidogly’can unit) accumulates in cells of Staphylococcus aureus treated with low concentrations of penicillin G or with ampicillin, methicillin, or cephalothin. The uncross-linked monomer has been isolated, and analyses indicate that it represents a prefabricated subunit of the wall bearing both of the o-alanine residues of its pentapeptide precursor as wel...
